R882H mutations in the DNA methyltransferase DNMT3A are often observed in AML tumors. We show in this work that this mutation strongly alters DNMT3A's preferences for DNA methylation within certain flanking sequences. In addition, the mutation causes the tetramerization of DNMT3A to change, leading to an amplification of the effects of the mutation in cells. Our data can help to better understand the effect of the R882H mutation in tumor cells in the futur
