Clr4 is a protein methyltransferase known to methylate histone H3 at lysine 9. This modification is essential for heterochromatin formation in S. pombe. We investigated the substrate specificity of Clr4 using peptide arrays. Our data show, that Clr4 recognizes an R-K-SKRT-TCS-G sequence motif and it methylates novel non-histone substrates.
