Specificity of SUV39H2

Methylation of lysine residues in proteins is a very important modification with several essential biological roles. Therefore, mapping lysine methylation sites and identifying the responsible protein lysine methyltransferases (PKMTs) are important scientific challenges and many groups are working on these subjects. Recently, the methylation of histone H2AX K134 by SUV39H2 was reported and it was proposed to have an important role in DNA repair (Sone et al., Nat. Commun., 2014, 5:5691). In this paper, we report results of in vitro methylation reactions with H2AX protein and peptides, in which no H2AX methylation by SUV39H2 was detectable. Potential reasons for this discrepancy and it consequences are discussed.
